Lentivirus vectors are one of the most efficient vehicles for delivering genes into a broad spectrum of cells, unique in their ability to penetrate both dividing and non-dividing cells. Their ability to integrate with the host genome allows for long-term gene expression and analysis in target cell lines. Because of their versatility, lentiviral vectors have proven indispensable tools in the field of gene delivery and gene therapy, and are frequently used in gene therapy of hematopoietic stem cell disorders and for immunotherapy.
We manufacture 3rd generation vectors consistent with guidelines provided by the Public Health Agency of Canada. This means they meet the highest safety standards on the market. They’re produced by transfection of HEK293T cells with 4 endotoxin-free expression plasmids
Furthermore, we provide QC Analytics services for preclinical and clinical lentivirus vectors. This service includes vector titration, vector copy number analysis and expression profile of gene of interest
Customizable
Our 3rd generation lentivirus vectors are modular, allowing the use of specific promoters (CMV, pGK, EF1-a, EFs) with specific markers (eGFP, mCherry, lacZ, PuroR, mPlum, mNeptune). We offer single and dual promoter vectors; the latter allows for expression of two genes simultaneously as well as inducible and CRISPR vectors. All vectors carry the vesicular stomatitis virus-G (VSV-G) envelope protein, known to target a wide variety of cell types. We also offer vector titration, vector copy number and transgene expression analysis services for preclinical and clinical grade lentivirus vectors.
High Titer and High Purity
Our vectors undergo a stringent ultrafiltration process, resulting in titers of 10e8 TU/mL and 10e9 TU/mL with purity that is near pre-clinical and clinical grade vectors. This guarantees highly efficient transfer of your gene into target cells (hematopoietic stem cells, primary cells, induced pluripotent stem cells, organoids), and facilitates gene expression analysis.
Custom LVV Kits
For researchers wishing to use their own gene of interest (GOI), we now have the “all-in-one” LV vector-Custom. With this kit, researchers simply add their 3rd Gen. transgene vector to the vector mix, which is then immediately added to producer cells, and LVV vector-Custom is harvested 48 hours later. Our LVV-Custom kits will produce up to 10-fold higher titer of LVV than other LVV kits on the market (NOTE: Titers can vary depending on the size and nature of the custom vector added).
All-In-One LVV Kits
Our new one-of-a-kind “all-in-one” LVV kits allows the researcher to produce high titer 3rd Gen LVVs in their own laboratories. Our kits contain all the expression plasmids in optimized ratio, together with the transfection reagent, to produce high titer LVV. Simply add the vector mix to producer cells (HEK293T) and harvest high titer LVV after 48 hours. In our LVV-eGFP kit, one vial allows production of greater than 10e8 total infectious units of LVV eGFP from a single 100mm plate. This kit is also available to produce LVV-BFP, LVV-mPlum, LVV-mNeptune or LVV-firefly luciferase.
Pre-made Lentivirus vectors
Tailored Genes provides ready-to-use lentivirus vectors (LVVs). These include LVV eGFP, LVV BFP, LVV mPlum, LVV mNeptune and LVV firefly luciferase. These vectors are available in 100uL aliquots at titers of 10e8 infectious units/mL. All vectors contain the EF1 alpha promoter, but we can design custom vectors with CMV, hPGK or EFs promoters to meet your individual needs.
